Model of KRAB-ZFP binding to DNA and induction of heterochromatin formation. Protein–DNA interaction between ZNFs and DNA are mainly mediated by four amino acids at positions −1, 2, 3, and 6 of the α-helix (colored circles). KAP1 is recruited through the KRAB domain and interacts with the NURD/HDAC repressor complex and histone methyltransferases (HMTs) (e.g. SETDB1), which catalyze the removal of H3K9ac and the addition of H3K9me3, respectively.
