Ovarian somatic piRNA pathway in Drosophila. After being spliced and transferred to nuclear Dot COM, flam transcripts are exported to cytoplasmic Yb bodies, believed to be the site of piRNA processing. Their processing in piRNAs requires 5′ end cleavage, loading onto Piwi protein and 3′ end trimming. The mature Piwi pi-RISC is then imported into the nucleus to silence target transposons by Transcriptional Gene Silencing (TGS).
